Nació en Chicago (Illinois) el 26 de octubre de 1912 y realizó sus estudios en Inglaterra en la Universidad de Cambridge. Su carrera cinematográfica comenzó en 1933 como bibliotecario en la Warner Bros. Tras algunos cortometrajes a principios de la década de 1940, en 1946 rodó su primera película The Verdict (con Peter Lorre). El estilo de sus películas, artesanalmente perfecto y carente de toda pretensión, como puede apreciarse en La invasión de los ladrones de cuerpos (1956) o Código del hampa (1964), entusiasmó sobre todo a los críticos de Cahiers du Cinéma. Más tarde se consagró como especialista en películas del oeste, trepidantes y llenas de tensión, así como en filmes de acción y policiacos. La jungla humana (1968) marcó el inicio de una colaboración enormemente fructífera entre Siegel y Clint Eastwood. La película policiaca Harry el sucio (1971), en la que Eastwood interpreta un agente carente de escrúpulos que alcanza su objetivo tomándose la justicia por su mano, resultó todo un éxito de taquilla. Don Siegel falleció el 20 de abril de 1991 en Nipomo (California).
